Luke signed for Harlequins RL from Manly Sea Eagles in September 2008. The son of former Queensland player Henry Williamson, Luke played his junior football for the Brothers Holy Spirit Hornets in Brisbane. In 1997 Luke graduated to the Australian Super League, when signed a first grade contract with the now defunct Adelaide Rams. In 1999 Luke moved on to Canberra for a two year period, before moving on to the Manly club in Sydney (at that time called Northern Eagles).

An accomplished goal-kicker, Luke played much of his early rugby at centre or half-back, before settling into the vacant loose forward position in 2006, when former Australia Test Player Ben Kennedy departed the club. Luke is a highly versatile and experienced player who is considered to be one of the top defenders in the game. A hard-working and consistent forward, Luke was also regarded at Manly as an excellent ambassador, both for the game and his club.

In 2007 Luke appeared for the Sea Eagles in the Grand Final defeat against Melbourne, before an 81,000 crowd at the Telstra Stadium, Sydney. Unfortunately for Luke, a late season hamstring injury caused him to miss a second Grand Final appearance, when Manly gained revenge over the Melbourne club.

Shortly after his arrival at Quins, Luke faced his former club, Manly, in the London Rugby League Challenge. Luke went on to recieve regular praise for his performances in his first Super League season and also brought with him invaluable experience to the side.
